Out-of-Equilibrium Measurements of Kinetic Constants on a Biosensor

Conventional measurements of kinetic constants currently in use are performed at equilibrium and may require large volumes, especially at low association rate constant k on. If the measurements are made out of equilibrium, the values obtained may be…

Endogenous Bos taurus RECQL is predominantly monomeric and more active than oligomers

There is broad consensus that RecQ family helicase is a high-order oligomer that dissociates into a dimer upon ATP binding. This conclusion is based mainly on studies of highly purified recombinant proteins, and the oligomeric states of RecQ…

Threshold accumulation of a constitutive protein explains E. coli cell-division behavior in nutrient upshifts

Despite a boost of recent progress in dynamic single-cell measurements and analyses in Escherichia coli , we still lack a mechanistic understanding of the determinants of the decision to divide. Specifically, the debate is open regarding the…

Structural and functional studies of SF1B Pif1 from Thermus oshimai reveal dimerization-induced helicase inhibition

Pif1 is an SF1B helicase that is evolutionarily conserved from bacteria to humans and plays multiple roles in maintaining genome stability in both nucleus and mitochondria. Though highly conserved, Pif1 family harbors a large mechanistic diversity…

Replication protein A plays multifaceted roles complementary to specialized helicases in processing G-quadruplex DNA

G-quadruplexes (G4s) are non-canonical DNA structures with critical roles in DNA metabolisms. To resolve those structures that can cause replication fork stalling and genomic instability, single-stranded DNA-binding proteins and helicases are…

Concerted conformational dynamics and water movements in the ghrelin G protein-coupled receptor

There is increasing support for water molecules playing a role in signal propagation through G protein-coupled receptors (GPCRs). However, exploration of the hydration features of GPCRs is still in its infancy. Here, we combined site-specific…

Modulation of cellular fate of vinyl triarylamines through structural fine tuning: to stay or not to stay in the mitochondria?

Mitochondria is involved in many cellular pathways and dysfunctional mitochondria are linked to various diseases. Hence efforts have been driven to design mitochondria-targeted fluorophores for monitoring the mitochondria status. However, the…
