switchSENSE – a biophysical tool for characterization of polymerases and antibodies

switchSENSE is a novel dynamic biosensing technique based on electro-switchable biosurfaces (ESB). The sensor employs DNA nanolevers, moving in an alternating electrical field to determine the hydrodynamic diameter (DH) of proteins and formed protein complexes. This gives insight into binding kinetics (kon, koff, KD) of complex molecular interactions of antibodies and their respective antigens (affinity & avidity). Moreover, direct attachment and modification of DNA nanostructures by DNA modifying enzymes such as polymerases, ligases as well as restriction enzymes is feasible. Elongation rates, ligation reactions and exonuclease activity can be monitored in real time. The principle will be explained accompanied by relevant examples.
